SAMMENFATNING: De rapporterede data fastslår, at behandling med 4 mg dextrothyroxine eller 0,15 mg levothyroxin af hypothyroide patienter resulteter i lignende grad af sænknin af serum TSH, kolesterol, triglycerider og phospholipid niveauer og stimulering af stofskiftet.

Murphy-Pattee (metide/red.) total T4 bestemmelse anvendes på blodprøver udtaget 24 timer efter den sidste dosis af dextrothyroxine kan bruges til at vurdere tilstrækkeligheden af behandlingen. Korrektion af hypothyreose kræver højere serumniveauer af dextrothyroxine end af levothyroxin.

Serum T3 niveauerne stiger hos patienter behandlet med dextrothyroxine. I behandlingen af hypothyreose virker dextrothyroxine lige godt på både sænkning af kolesterol og stigning af stofskifte. Yderligere undersøgelser er nødvendige for at kunne fastslå, om en sådan effekt kan påvises i euthyroid hypercholesterolemic forsøgspersoner med doser fastsat heri, som ligestillede med hensyn til den stimulerende virkning på stofskiftet.

The effect of d-thyroxine on the metabolism of cholesterol in familial hyperbetalipoproteinaemia

Abscract: The effect of d-thyroxine plus propranolol on cholesterol metabolism was investigated in three patients with familial hyperbetalipoproteinaemia whose plasma cholesterol was labelled with [14C]cholesterol. In each patient the treatment caused a fall in plasma cholesterol concentration and a temporary increase in faecal excretion of endogenous steroids, due to increased output of neutral steroids in two patients and to increased output of bile acids in the third patient. The cumulative increase in output of endogenous steroids during treatment was greater than the amount of cholesterol lost from the plasma. Since the specific radioactivity curve of the plasma cholesterol did not indicate increased synthesis of cholesterol during the treatment, it was concluded that cholesterol mobilized from the tissues, as well as plasma cholesterol, contributed to the increase in faecal excretion of endogenous steroids.
